Y analytical pro tein assay metnoas in rats witn surrace glomerun, we measured glomerular capillary and proximal tubule hydrostatic pressures and estimated afferent and efferent arteriolar oncotic pressures during conditions deliberately designed to increase GFR (acute expansion of plasma and extracellular volume). Single-nephron (SN) GFR and initial glomerular capillary plasma flow (GPF) both increased approximately twofold. Net ultrafiltration pressure at the afferentmost point in the glomerulus was essentially the same prior to and following volume expansion. Similarly, filtration pressure equilibrium was achieved both prior to and following expansion. The roughly proportional changes in SNGFR and GPF provide evidence that glomerular ultrafiltration is a plasma-flow-dependent process. Under these conditions (filtration pressure equilibrium), SNGFR is determined entirely by GPF, transcapillary hydrostatic pressure difference, and initial capillary protein concentration. The possible physical mechanisms underlying the plasma-flow dependence of SNGFR have been examined using a recently developed model. along the glomerular capillary (8). We report here of measurements related to the dynamics of gloultrafiltration during one such experimental mathat of deliberately elevating GFR by means of acute expansion of plasma and extracellular volume. The results of these measurements provide evidence to indicate that glomerular ultrafiltration is a plasma-flow-dependent process. Using these measurements and the relationships derived from the mathematical model (8), it has been possible to examine the relative influences of several factors potentially responsible for this phenomenon of flow dependence.

Dynamics of glomerular ultrafiltration in the rat. IV. Determination of the ultrafiltration coefficient.

Pressures and flow rates were measured in accessible surface glomeruli of mutant Wistar rats under conditions deliberately designed to prevent achievement of filtration pressure equilibrium, that is, the equalization of transcapillary hydrostatic and oncotic pressures by the efferent end of the glomerulus as typically observed in the normal hydropenic rat. Disequilibrium was obtained at elevate...

Modulation by prostaglandin synthesis inhibitors of the action of exogenous angiotensin II on glomerular ultrafiltration in the rat.

We studied the effects of angiotensin II (A II) infusion, with and without inhibition of prostaglandin (PG) synthesis, on the determinants of glomerular ultrafiltration in 27 plasma volume-expanded Munich-Wistar rats. The effects of PG inhibition alone and infusion of vehicle alone also were investigated. With a pressor dose of A II single nephron (SN) GFR and total GFR did not change significa...

Converting enzyme inhibition and the glomerular hemodynamic response to glycine in diabetic rats.

GFR normally increases during glycine infusion. This response is absent in humans and rats with established diabetes mellitus. In diabetic patients, angiotensin-converting enzyme inhibition (ACEI) restores the effect of glycine on GFR. To ascertain the glomerular hemodynamic basis for this effect of ACEI, micropuncture studies were performed in male Wistar-Froemter rats after 5 to 6 wk of insul...

Renal Function and Hemodynamic Study in Obese Zucker Rats

OBJECTIVES To investigate the renal function and hemodynamic changes in obesity and hyperinsulinemia which are characteristics of type II diabetes. METHODS Studies were carried out in two groups of female Zucker rats. Group 1 rats were obese Zucker rats with hereditary insulin resistance. Group 2 rats were lean Zucker rats and served as controls. In comparison with lean Zucker rats, obese Zuc...

The dynamics of glomerular filtration in the puerperium.

We evaluated the glomerular filtration rate (GFR) during the second postpartum week in 22 healthy women who had completed an uncomplicated pregnancy. We used physiological techniques to measure GFR, renal plasma flow, and oncotic pressure and computed a value for the two-kidney ultrafiltration coefficient (K(f)). We compared these findings with those in pregnant women previously studied on the ...
